The estimation of urobilinogen is an important diagnostic test used to assess liver function and detect certain medical conditions. Aim:-
Estimation of Urobilinogen test by Ehrlich method.
Principle:-
Urobilinogen react with P – dimethyl amino benzaldehyde in
acidic medium (in Ehrlich reagent) to form pink color compound.
Requirement:-
Centrifuge, test tube, pipette, 10 g/dl Barium chloride,
Ehrlich reagent.
Procedure:-
1. Place 3 to 4 ml urine in a centrifuge tube by using a pipette.
2. Add equal amount of 10 g/dl Barium chloride and mix well.
3. Centrifuge at 1500 RPM for 10 minutes and place supernent another tube.
4. Add about 0.5 ml of Ehrlich reagent to supernent.
Observation:-
Development of pale pink color Urobilinogen present normal while development of cherry red color Urobilinogen presents high.
